Its the mathematical modeling of the drugs path through the body.  ie; once
taken, either by IV route directly into the bloodstream or by other routes
where the drug must be liberated from its dosage form, absorbed into the
blood stream, distribution into body compartments (ie in a manner one can
use for mathematical modeling) metabolism , (including first pass through
the liver when taken orally), and elimination.
