How to make Provenge

In case you want to make it at home, here's what Provenge
("Sipuleucel-T") is and how to make it.

   "Sipuleucel-T is a vaccine product consisting of autologous
   DCs pulsed ex vivo with a recombinant fusion protein
   (PA2024), consisting of full-length PAP linked via its COOH
   terminus to the NH2 terminus of full-length granulocyte
   macrophage colony-stimulating factor (GM-CSF). PAP is an
   antigen expressed solely in prostate tissue and in 95% of
   prostate cancer cells, thus serving as an ideal target for
   vaccine-based therapy [18,19]. GM-CSF acts as a targeting
   molecule that directs the PAP antigen to DCs and promotes
   antigen uptake and processing.  Preparation of sipuleucel-T
   involves leukopheresing 1-2 blood volumes (8-14 l) with
   isolation of peripheral blood mononuclear cells (PBMCs) via
   two centrifugations over a buoyant density solution of 1.0770
   g/ml and 320, 1.0650 and 320 mOsm, respectively, to allow
   removal of platelets and monocytes. The cell pellet, which
   contains DC precursors, is washed and incubated for 40 h in
   AIM media with 10 µg/ml of PA2024 in the absence of
   exogeneous cytokines or serum. After incubation, the cells
   are washed and suspended in lactated Ringer's solution for
   infusion. Sipuleucel-T is currently administered three times;
   once every 2 weeks."  (from "Expert Review of Cancer Therapy"

I tried to do it that way, but it didn't work.
