Sodium ascorbate (vitamin C) induces apoptosis in melanoma cells via the down-regulation of transferrin receptor dependent iron uptake.

Sodium ascorbate (vitamin C) has a reputation for inconsistent effects upon malignant tumor cells, which vary from growth stimulation to apoptosis induction. Melanoma cells were found to be more susceptible to vitamin C toxicity than any other tumor cells. The present study has shown that sodium ascorbate decreases cellular iron uptake by melanoma cells in a dose- and time-dependent fashion, indicating that intracellular iron levels may be a critical factor in sodium ascorbate-induced apoptosis. Indeed, sodium ascorbate-induced apoptosis is enhanced by the iron chelator, desferrioxamine (DFO) while it is inhibited by the iron donor, ferric ammonium citrate (FAC). Moreover, the inhibitory effects of sodium ascorbate on intracellular iron levels are blocked by addition of transferrin, suggesting that transferrin receptor (TfR) dependent pathway of iron uptake may be regulated by sodium ascorbate. Cells exposed to sodium ascorbate demonstrated down-regulation of TfR expression and this precedes sodium ascorbate-induced apoptosis. Taken together, sodium ascorbate-mediated apoptosis appears to be initiated by a reduction of TfR expression, resulting in a down-regulation of iron uptake followed by an induction of apoptosis. This study demonstrates the specific mechanism of sodium ascorbate-induced apoptosis and these findings support future clinical trial of sodium ascorbate in the prevention of human melanoma relapse. One notices the addition of deferoxamine .. ADDS to the .. killing ...
Therefore one might surmise .. iron reduction .. to the POINT .. of .. the body itself BEING .. 'able' to sequester / bindup the iron .. itself .. might be worth the effort .. ?
That would mean lowering the iron levels down to the level found in a normal person which would be the 'new hypothesis' .. of 25% transferrin saturation ..

Iron: a target for the management of Kaposi's sarcoma?

BACKGROUND: Kaposi's sarcoma (KS) is a mesenchymal tumour associated with human herpesvirus-8 infection. However, the incidence of human herpesvirus-8 infection is far higher than the prevalence of KS, suggesting that viral infection per se is not sufficient for the development of malignancy and that one or more additional cofactors are required. DISCUSSION: Epidemiological data suggest that iron may be one of the cofactors involved in the pathogenesis of KS. Iron is a well-known carcinogen and may favour KS growth through several pathways. Based on the apoptotic and antiproliferative effect of iron chelation on KS cells, it is suggested that iron withdrawal strategies could be developed for the management of KS. Studies using potent iron chelators in suitable KS animal models are critical to evaluate whether iron deprivation may be a useful anti-KS strategy. SUMMARY: It is suggested that iron may be one of non-viral co-factors involved of KS pathogenesis and that iron withdrawal strategies might interfere with tumour growth in patients with KS.

It is never a good idea to post your real name and personal information on the Internet. It can be used by unscrupulous individuals to harass you. Some trolls and lunatics will post your email address all over the Internet if you disagree with them. They are also known to add you to spam lists or sign you up to porn subscriptions. They have also forged emails and posted to sexually explicit newsgroups using their targets address. This is intended to embarrass the target or get them in trouble with the law.
I never use real information online. I also change my address when it appears that spammers have me on their lists. Look into aliases, they can be changed in a few minutes and do not require a call to your ISP. If that does not work look into a throw away email account from a free service like Hotmail.
Change your address as often as needed. It is your call to decide how often that might be.
Do not use your name in your email address, if that address will be posted on newsgroups. Also, if you need to post your email address, be sure to munge it or spam proof it. You can explain in the text how to make it usable.
