Plasma paraoxonase activities, lipoprotein oxidation, and trace element
interaction in asthmatic patients.

Paraoxonase (PON1) protects low and high-density lipoproteins (LDL and
HDL) against oxidation induced by reactive oxygen species formation
facilitated by iron (Fe) and copper (Cu) ions. Plasma PON1,
arylesterase, oxidized LDL (Ox-LDL), Cu, Fe, thiobarbituric
acid-reactive substances (TBARS), lipid, lipoprotein, and
apolipoprotein profile in bronchial asthma were determined and the
relations among these parameters in different steps of asthma were
interpreted. A total of 58 individuals, 30 asthmatics and 28 controls,
were included into the scope of this study. Plasma PON1, arylesterase,
and TBARS levels were measured spectrophotometrically. Determination of
plasma oxidized LDL, Cu, and Fe levels were performed by enzyme-linked
immunosorbent assay, atomic absorption spectrophotometry, and the
automated TPTZ method, respectively. Apo-A-1 and Apo-B levels were
determined immunoturbidometrically. Plasma total cholesterol,
triglyceride, and HDL cholesterol levels were enzymatically determined.
Plasma LDL levels were estimated using the Fridewald formula. The
average plasma PON1 and arylesterase activities in the group of
patients were lower than those of the individuals in the control group,
but there was no statistically significant difference found between
them (p > 0.05). No significant difference was found in plasma Apo-A-1,
Apo-B, total cholesterol, triglyceride, HDL, and LDL concentrations
between the control and patient groups (p > 0.05). Plasma oxidized LDL
(p < 0.05), Cu (p < 0.01), Fe (p < 0.01), and TBARS (p < 0.001) levels
in patients with asthma were found to be significantly higher than for
the control group. Increases in Cu, Fe, lipid peroxidation, and
oxidized LDL levels supported by relative decreases in PON1 activities
observed in asthmatic patients might be introduced as the striking
findings as well as the possible potential indicators of this airway
disease, the prevalence of which has increased dramatically over recent
decades.

Copper {Cu] and Iron {Fe] .. are oxidizers .. they destroy antioxidants

Lipid peroxidation

Definition:

 The process whereby free radicals "steal" electrons from the
lipids in our cell membranes, resulting in cell damage and increased
production of free radicals.

Oxidizers such as .. copper .. and .. iron .. cause free radicals ..

PON1 is an antioxidant .. and it is .. depleted ..

All of the above point to oxidation .. BEING .. 'present' .. IN ..
'those with asthma' ..

Sooooo .. they are saying .. "we believe these in PARTICULAR .. copper
.. iron .. lipid peroxidation .. and decreased PON1 .. can be used to
TELL {potentially indicate] whether or not one 'has' / diagnosis ..
asthma .." ..

'Striking' means ..  to hit .. to picket against a perceived fairness
inequality or in this .. case .. "holy sht" ..

IMHO ..

look what does that mean? That people are not taking or getting adequate
amounts of iron and this is precipitating more asthma? Seriously I am not
doubting that as a reason but certainly I have my own reasons. Go to alt
lawyer. for why I think pollution is the single biggest reason for adult
onset asthma and probably illness in general in the US except for maybe
smoking and diet...

Totally irrelevant question to ask a crackpot who thinks all disease is
caused by iron!

Lactoferrin is an .. iron binder .. and it is produced in our bodies ..
and when this protein is 'overloaded' .. saturated with iron .. it
cannot do its' .. job ..

Sooo .. is the 'treatment' .. 'emptying' our naturally occuring /
produced lactoferrin .. so it can NOW .. do its' .. job .. ?

When INTRODUCED .. it seems it is .. effective in **reducing** ..
asthma and allergy .. symptoms ..

Pollen grains contain reduced nicotinamide adenine dinucleotide
phosphate (NAD(P)H) oxidases and in contact with mucosal surfaces
generate superoxide anion (O(2) (*) (-)). In the presence of iron, O(2)

(*-) may be converted to more reactive oxygen radicals, such as to
H(2)O(2) and/or (*)OH, which may augment antigen-induced airway
inflammation. The aim of the study was to examine the impact of
lactoferrin (LF), an iron-binding protein, on ragweed (Ambrosia
artemisiifolia) pollen extract (RWE)-induced cellular oxidative stress
levels in cultured bronchial epithelial cells and accumulation of
inflammatory and mucin-producing cells in airways in a mouse model of
allergic airway inflammation. Results show that LF lowered RWE-induced
increase in cellular reactive oxygen species (ROS) levels in bronchial
epithelial cells. Most importantly, LF significantly decreased
accumulation of eosinophils into airways and subepithelium of
intranasally challenged, sensitized mice. LF also prevented development

of mucin-producing cells. Amb a 1, the major allergenic ragweed pollen
antigen lacking NAD(P)H oxidase activity, induced low-grade airway
inflammation. When administered along with glucose oxidase (G-ox), a
superoxide-generating enzyme, Amb a 1 induced robust airway
inflammation, which was significantly lowered by LF. Surprisingly, LF
decreased also inflammation caused by Amb a 1 alone. Iron-saturated
hololactoferrin had only a marginal effect on RWE-induced cellular ROS
levels and RWE- or Amb a 1 plus G-ox-induced inflammation. We postulate

that free iron in the airways chemically reduces O(2) (*) (-) to more
reactive species which augment antigen-induced inflammation in a mouse
model of asthma. Our results suggest the utility of LF in human
allergic inflammatory disorders.
