"Chlorogenic acids and the flavonols"

Isolation and Structure Elucidation of Antioxidant Polyphenols from
Quince (Cydonia vulgaris) Peels. [JOURNAL ARTICLE]

Thirteen new compounds, as well as 16 already known, have been
isolated from organic extracts of peels of Cydonia vulgaris, a fruit
of a shrub belonging to the same tribe as the apple. All of the
structures were elucidated by EI- or ESI-MS and (1)H and (13)C NMR
after purification of individual compounds by HPLC.
Thirteen fatty acid esters of cinnamyl alcohols, three fatty acid
esters of hydroxybenzoic acid, three fatty acid esters of
hydroxybenzaldehyde, three glucosides of aromatic acids, four
chlorogenic acids, two flavonols, and a benzylamine have been
identified.
The fatty acid moieties have been identified by GC-MS analysis of the
methanolysis products. All of the compounds were tested for their
radical scavenging and antioxidant activities by measuring their
capacity to scavenge the 2,2'-diphenyl-1-picrylhydrazyl (DPPH) radical
and anion superoxide radical and to induce the reduction of Mo(VI) to
Mo(V).
The chlorogenic acids and the flavonols exhibited more antioxidant and
radical scavenger capacity than the positive standards alpha-
tocopherol and ascorbic acid.
The results of the tests were analyzed by cluster analysis that
grouped all of the compounds on the basis of the substituents on the
aromatic ring.

mitogen-activated protein kinase-dependent apoptosis in chronic
myelogenous
leukemic cells.

We report that chlorogenic acid (Chl) induces apoptosis of several
Bcr-Abl-positive chronic myelogenous leukemia (CML) cell lines and
primary
cells from CML patients in vitro and destroys Bcr-Abl-positive K562
cells in
vivo. In contrast, this compound has no effect on the growth and
viability of
Bcr-Abl-negative lymphocytic and myeloid cell lines and primary CML
cells.
Sodium chlorogenate (NaChl) exhibits 2-fold higher efficiency in
killing K562
cells compared with Chl. NaChl also induces growth inhibition of
squamous cell
carcinoma (HSC-2) and salivary gland tumor cells (HSG), although at 50-
fold
higher concentration. NaChl inhibits autophosphorylation of p210(Bcr-
Abl)
fusion protein rapidly. We demonstrate that p38 phosphorylation is
increased in
Bcr-Abl-positive cells after treatment with NaChl and closely
paralleled the
inhibition of Bcr-Abl phosphorylation. NaChl did not increase
phosphorylation
of p38 in Bcr-Abl-negative cells including HSC-2 and HSG that are
responsive to
this compound, indicating that p38 activation by NaChl is dependent on
Bcr-Abl
kinase inhibition. Inhibition of p38 activity by SB203580
significantly reduced
NaChl-induced apoptosis of K562 cells, whereas activation of p38 by
anisomycin
augmented the apoptosis. These findings indicate that inhibition of
Bcr-Abl
kinase leading to activation of p38 mitogen-activated protein (MAP)
kinase may
play an important role in the anti-CML activity of Chl.

and muscle fatigue: Of ROS, canaries, and the IOC,' have been
published.
According to a study from the United States, "Skeletal muscle fibers
continually generate reactive oxygen species (ROS) at a slow rate
that
increases during muscle contraction.
This activity-dependent increase in ROS production contributes to
fatigue of skeletal muscle during strenuous exercise."
"Existing data suggest that muscle-derived ROS primarily act on
myofibrillar proteins to inhibit calcium sensitivity and depress
force.
Decrements in calcium sensitivity and force are acutely reversible by
dithiothreitol, a thiol-selective reducing agent.
These observations suggest that thiol modifications on one or more
regulatory proteins are responsible for oxidant-induced losses during
fatigue.
More intense ROS exposure leads to losses in calcium regulation that
mimic pathologic changes and are not reversible.
Studies in humans, quadrupeds, and isolated muscle preparations
indicate that antioxidant pretreatment can delay muscle fatigue.
In humans, this phenomenon is best defined for N-acetylcysteine
(NAC),
a reduced thiol donor that supports glutathione resynthesis. NAC has
been shown to inhibit fatigue in healthy adults during electrical
muscle activation, inspiratory resistive loading, handgrip exercise,
and intense cycling," wrote M.B Reid and colleagues, University of
Kentucky, Department of Physiology.

The researchers concluded: "These findings identify ROS as endogenous
mediators of muscle fatigue and highlight the importance of future
research to (a) define the cellular mechanism of ROS action and (b)
develop antioxidants as novel therapeutic interventions for treating
fatigue."

Reid and colleagues published their study in Free Radical Biology &
Medicine (Free radicals and muscle fatigue: Of ROS, canaries, and the

I believe they're right on target about the problem, and I hope they're
right about the cure, except for the "irreversible" bit!

But it isn't even March 30 yet, and I can't find "Kentucky" or "radical"
at newsrx.com.  Where'd you find that?

too.

I use individual.net and I lOVE it. Don't see the crazy spam, posts show
up really quickly, etc etc. I used it when it was free now there's a
nominal 10 EU charge. They are great and it's a pleasure for me to give
them a few bucks/year. Funny to look up the exchange rate... Anyway it's
easy and works well...
