Howard Shapiro's PFC 2nd edition answers this (perhaps just in
passing, if I
remember it rightly - someone permanently "borrowed" my copy long
ago):
Pre-Computer-driven cytometers,  used multichannel analysers (MCA's)
to
acquire and display data - these record pulse heights and plot the
statistical frequency of pulses of different heights and so were used
on
cytometers to generate and display single-parameter intensity
histograms.

Before being hijacked by cytometrists however, MCA's were originally
designed for and used in nuclear (gamma) spectroscopy, where the
height of
the detector pulse would be related to the energy (and thus spectral
frequency) of the single photon generating it, so originally each
"channel"
recorded the statistical frequency of photons having a narrow range of
spectral frequencies - spectral frequency range is, as you point out
in your
query, one of  the prevalent uses of the word "channel" .

The "channel" nomenclature seems to have stuck even when the pulses
measured
corresponded to the intensity of monochromatic (near as dammit)multi-
photon
pulses who's height corresponded not to spectral position, but to
marker
density on particles - and it has done so quite stably if mystifyingly
over
quite a time now that ADC's feed our PC's the information ! :)

This brings up a question I've idly wondered about for some time.

Flow cytometry seems to be unique in the sense that "channel" refers
to the
specific output level of an A/D converter which is related, in turn,
to the
number of photons measured by a PMT.

The more prevalent use of the term "channel" I've seen is in reference
to a
defined band of electromagnetic energy that generally has some finite
width,
such as a range of light wavelengths or radio frequencies used
together as a
communication band. In non-flow cytometric fluorescence detection
systems,
I've always seen it used to refer specifically to a color channel or
spectral band. Confusingly, I've seen "channel" used in both senses in
flow
(referring to an A/D output level or to a spectral detection band).

Does anybody know the historical reason for the use of "channel"
terminology
for A/D output in flow cytometry?

To be even more scientific:

"In physics, intensity is a measure of the time-averaged energy flux.
The
word "intensity" here is not synonymous with "strength", "amplitude",
or
"level", as it sometimes is in colloquial speech" /Wikipedia,
Intensity (as
much as Wikipedia is scientific)/

The units of electromagnetic wave, or light intensity in SI are W/m²
(watts
per square meter). In our case we are talking about channel numbers.
They do
represent intensity, but the numbers that we see are numbers assigned
to
channels, hence MFC, not MFI.

I often see in papers fluorescence "intensity" graphs with the axis
labeled
MFI, but I always label my graphs MFC, which is what the axis units
actually
are - channel numbers (whether mean or median).

Since MFI is not a "generally accepted" abbreviation (for journals),
it has
to be defined anyway in manuscripts.

Given that it could refer to either statistic, it should always be
defined
even in less formal settings (e.g., EMail).  That being said, I would
hazard
a guess that "mean" is far more commonly-used and most people will
assume
"mean" if not explicitly defined otherwise.

To be safe.... always be explicit on the first use.

someone do a check on them. it is apparent that they have written

software and they think its ants-pants. They have tried to post

on the Purdue list, they have used every possible website and email

discussion group to get cross listed to boost their ratings on
Google.

Basically I view their behavior as very tenuous and from the message

sent me, it appears that it is not appropriate to do what they are
doing...

I have taken the following action:

1. Steve Kelley has been instructed to remove their email and any

postings on the Purdue list. They are now banned

2. I am going to Copying Bartek Rajwa the editor of the ISAC site to

beware of them

3. I am contacting Google and other sites to let
them know that these

people are self-propagating links.

4. If I see any message that in any way impunes Purdue or our

reputation, I will go after them with every possible legal recourse
at

my disposal. We will not allow our reputation to suffer because of

commercial abuse.

5. You should ban this address, and basically indicate to your
members

that this is a scam. While they claim to be legitimate, they are
acting

exactly as any scam artist...I already told them that they were
acting

as scammers and they got upset with me...well, if they don't desist,

they will find out how much influence we actually have...
